DNA Preparation Plasmid DNA/phage DNA as well as PCR fragments can be used as template. It is essential to submit your DNA in water (without EDTA or salts), with an accurate estimated DNA concentration (> 0.1ug/ul), as we will not confirm this. Please note if the template is > 8kb, since increased amounts of DNA will be needed for each reaction. For PCR fragments, the minimum amount of template DNA is ~1ng/bp (i.e. 150ng for a 150bp fragment).
Primer Preparation Dilute primers in water to a concentration of 3-6 uM. It is best to design primers to have a Tm of 48-54°C, to end with a 3' C or G, and be ~20 nucleotides. If using the nominal approximation, Tm = [(4) (G + C) + (2) (T + A)], the "Tm" must be > 60°C. Please note that we provide many primers free of charge (M13F/R, T3, T7, T7 term, pGEX F/R, Sp6, BGH, etc.).
Related Services To ensure reproducible sequencing results, we provide rapid DNA isolation / purification services to enable sequencing directly from bacteria, yeast, phage and PCR reactions. We also design and synthesize primers, using programs to minimize primer dimers and hairpin loops, to facilitate projects requiring complete sequencing of cloned insert. Quality Assurance A control sequencing reaction is carried out with each order. Should the control reaction fail, then all sequences will be rerun at no cost. In our experience, the success of each reaction is highly dependent on the concentration / purity of the submitted DNA and primer . In projects where we are responsible both for making the DNA and designing the primer, your sequencing results are guaranteed.
How to order Please complete the DNA Sequencing Order Sheet listing the template and primer identifications exactly as written on the sample tubes, and indicate which output format you prefer (PC or Mac). Each of your sequences will be assigned a unique Veritas sequence ID number for future reference. Send template materials and primers to the address below. Be sure to include the sequencing order sheet to avoid any delays. For customers in the Rockville/Bethesda area (including NIH) we also offer "same-day" courier pick up service for a nominal charge ($12.00)
* Please contact us about academic / institutional discounts
** If a "fast" edit of each reaction is required (up to 500 nt) please add $8 per reaction.
